<p>Replacing of harmful organic solvents such as hexane with a safe and environmentally friendly green method, i.e., enzymatic process in aqueous phase for extracting oil from oilseeds, is essential for consumer health and preventing environmental hazards. In this regard, combined carbohydrases and proteases were used in a single-step aqueous phase process for oil and protein hydrolysates extraction from sesame seeds. The physicochemical characteristics of sesame seeds showed 24.16% protein, 54.19% fat, 2.58% moisture, 11.78% fiber, 15.98% total saturated fatty acids, 39.15% oleic acid, and 44.10% linoleic acid. At first, the optimum process conditions of Viscozyme L, Pectinex XXL, and Celluclast 1.5 L (carbohydrases) and Flavourzyme 1000 L, Alcalase 2.4 L, and Neutrase 0.8 L (proteases) were evaluated. Then, various enzyme mixture ratios were assessed to select the best multi-enzyme combination. The results revealed that 0.5–1.0% viscozyme, alcalase, and flavourzyme mixture with 1:1:1 ratio, in 2:1 ratio of water/sesame, at 55° C, pH 5.5, with 6-h holding time, was the optimal process condition. The combined enzymes have a t<sub>1/2</sub> up to 144&#xa0;min at 85&#xa0;°C. Kinetic assessment implied 2.44 × 10<sup>7</sup>–3.32 × 10<sup>8</sup> (M<sup>−1</sup>S<sup>−1</sup>) catalytic efficiency. Oil extraction yield was 93.73%. The protein content was enhanced 3 times after hydrolysis compared to raw sesame with 10–20&#xa0;kDa a molecular mass along with raising in essential amino acids content between 1.37 and 3.05 times compared to raw sesame protein and dried whole egg protein. Consequently, this process introduces itself an efficient, eco-friendly, and safe method for extraction of oil and protein hydrolysates from sesame seeds.&#xa0;</p>
