Background <p>Postmenopausal women have a higher prevalence of osteoporosis (OP) due to age related factors and hormonal changes. This study aims to investigate the interactions between lncH19 and miR-196a-5p, as well as their roles in the diagnosis and management of OP.</p> Aims <p>The ultimate goal is to identify novel biomarkers and potential therapeutic targets for this disease.</p> Methods <p>The relative expression levels of lncH19 and miR-196a-5p were measured using RT-qPCR. The diagnostic potential of lncH19 for OP was evaluated via receiver operating characteristic (ROC) curve analysis. The interaction between lncH19 and miR-196a-5p was verified by a luciferase reporter assay. To explore the regulatory effect of lncH19 in the pathogenesis of OP, PMA-induced THP-1 cells were used as an in vitro model.</p> Results <p>In postmenopausal women with OP, serum lncH19 levels were decreased, whereas miR-196a-5p levels were increased, and the two exhibited a negative correlation. LncH19 showed significant clinical diagnostic value for OP. It was confirmed that lncH19 directly targets miR-196a-5p and can suppress PMA induced inflammatory responses in macrophages. However, miR-196a-5p mimics could attenuate the anti-inflammatory effect of lncH19.</p> Conclusions <p>This study provides new insights into the pathogenesis of OP, clarifies the important value of lncH19 as a predictive biomarker, and offers a new direction for the treatment and prognostic evaluation of OP.</p>
