<p>Knowledge of the&#xa0;species diversity, relative abundance, and distribution pattern of various vector species of a particular region is significant in informing about disease transmission and implementing vector control strategies.&#xa0;The present study reports the prevalence of <i>Aedes lineatopennis</i> as a new mosquito record for Odisha, India. This finding is supported by both morphological and molecular analyses, specifically targeting the cytochrome oxidase c subunit I (COI) gene for the first time. The COI gene sequence of <i>Ae. lineatopennis</i> showed closeness from various populations across the globe showing the lowest genetic distance with Japanese (0.002) and highest with the Chinese (0.07) populations. The current study also presents a consolidated and updated record of mosquito species prevalent in India. The previously published checklist from 2015 documented 404 species. Since then, ten new mosquito species have been described from the country: <i>Hulecoeteomyia cherrapunjiensis</i>, <i>Uranotaenia (Pseudoficalbia) maikalensis</i>, <i>U. (P.) dandakaranyensis</i>, <i>U. (P.) satpuraensis</i>, <i>U. (P.) pseudostricklandi</i>, <i>Paraedes jambulingami</i>, <i>Lutzia (Metalutzia) tigripes</i>, <i>Culex (Culex) katezari</i>, <i>Cx. (Cx.) gaugleri</i>, and <i>Cx. (Cx.) kodaikanalensis.</i> This addition of new species has resulted in updating the tribes Aedini, Culicini, and Uranotaeniini within the subfamily Culicinae. Additionally, the study provides a concise overview of the larval habitats, taxonomic positions, distributions, and diagnostic characteristics of newly added mosquito species. This updated database would be helpful for the management of vector-borne diseases as well as the molecular diagnosis of <i>Ae. lineatopennis</i> would be significant in species confirmation for the other researchers.</p>
