Molecular authentication of Chironomidae larvae (Insecta: Diptera) using mitochondrial cytochrome oxidase-I gene sequence

Mitochondrial cytochrome oxidase subunit-I (COI) is one of the most applied DNA marker genes to the biodiversity, taxonomic and phylogenetic assessments. We have carried out morphological as well as DNA based identification and phylogenetic analysis of Chironomidae larvae. Morphological features of larval specimen were studied to evaluate species variation and COI gene was extracted and sequenced for phylogenetic analysis. The three species Chironomus circumdatus, Chironomus javanus, Polypedilum nubifer belongs to Chironominae subfamily and remaining Procladius sp. belongs to the subfamily Tanypodinae. Both genetic distance (K2P) and phylogenetic analyses unanimously confirmed identification. This molecular approach not only identified Chironomidae larval species precisely but also enhanced our understanding of their phylogenetic relationships and ecological roles.
