<p>Wild birds that are closely associated with humans can serve as reservoirs of zoonotic pathogens. The Village Weaver <i>Ploceus cucullatus</i> is a widely distributed passerine in sub-Saharan Africa and occurs in a wide range of ecological niches. Still, its gut microbiota and contribution to pathogen transmission are poorly understood. This study compares the gut microbiota of northern and southern Nigerian Village Weaver populations to determine regional variations and zoonotic potential. Fecal samples from both populations were analyzed using high-throughput 16S rRNA sequencing. Taxonomic assignments revealed the preponderance of members of the family <i>Enterobacteriaceae</i> (95.94%; <i>Enterobacter cloacae, Escherichia coli, Salmonella enterica, Klebsiella pneumoniae</i>) in northern gut microbiota, while <i>Campylobacteraceae</i> (70.30%; <i>Campylobacter lari</i>) and <i>Mycoplasmataceae</i> (28.17%; <i>Mycoplasma penetrans</i>) dominated the southern gut microbiota. Statistical analysis of the gut microbiota (north and south) revealed significant statistical differences (<i>p</i> &lt; 0.05) at all the hierarchical levels. Pathogenic strains of public health importance such as <i>Enterobacter cloacae</i> subspecies&#xa0;<i>cloacae</i> ATCC 13047, <i>Escherichia coli</i> O157:H7, <i>Escherichia coli</i> O104:H4, <i>Escherichia coli</i> O44:H18, <i>Escherichia coli</i> O145:H28, <i>Escherichia coli</i> O6:H16, and serovars of <i>Salmonella enterica</i> subspecies <i>enterica</i> (Newport, Havana, Typhimurium, Enteritidis, 1,4,[5],12:i:-, Indiana, Montevideo, Hvittingfoss, Heidelberg, Typhi) were recovered in the northern gut microbiota, while <i>Mycoplasma penetrans</i> was exclusively recovered from the southern gut microbiota. Findings from this study revealed that regiospecific adaptations and environmental conditions modulate and shape the gut microbiota of the Village Weaver. Owing to their broad feeding habits, the species could serve as conduits for effective transmission and dissemination of zoonotic pathogens via the fecal–oral route.</p>
