Phenotypic and genotypic resistance patterns of Escherichia coli in livestock from Dar es Salaam Tanzania

Misuse of antibiotics in livestock is a growing threat, but studies on antibiotic resistance in these animals, where interaction and antibiotic use are high, remain limited. A total of 155 E. coli isolates from pig, goat, and cattle cases were collected from Dar es Salaam abattoirs and farming regions. Antibiotic resistance against nine commonly used antibiotic agents was determined, and 12 drug-resistant genes were tested. Pigs showed the highest rates of antibiotic resistance: Ampicillin and chloramphenicol (50.9%), trimethoprim/sulfamethoxazole (49%), tetracycline (47%) and erythromycin (41.2%). Prevalence of resistance to the same five antibiotics in cattle was 34%, 26%, 44%, 58% and 32%. Slightly lower resistance rates were observed among isolates from goats, which were 51.8%, 20.4%, 22%, 40.7%, and 22%. In addition, pigs had the highest prevalence of multidrug resistant (MDR) isolates at 56%, followed by cattle at 35.2% and goats at 19.6%. The overall MDR prevalence among all livestock was 36.8%. Most resistance markers analysed were present in all but few isolates with the respective resistance at comparable rates. The markers analysed were the aadA1, blaTEM, blaCTX-M-1, catA1, cmlA, dfrA1, ere(A), qnrA, sul1, tet(A) and tet(B). There was lack of the ere(A) marker among isolates from pigs. The blaCTX-M-1 marker was not detected from any isolate. The study demonstrates the momentous contribution of pig keeping in the maintenance and spread of antibiotic resistance and multidrug resistance, a potential health risk to human. Expeditious interventions are imperative to protect human and animal health.
