Biodegradation efficacy of Exiguobacterium sp. HSK30 towards vegetable packaging LDPE film

The most popular packaging material is low density polyethylene (LDPE). The demand of LDPE has been increased due to its extensive properties which in turn producing more plastic waste and ultimately leads to plastic pollution. Recently, there has been a lot of focus on biodegradation of plastic because of the growing demand and strong need for its safe disposal. In the current study, bacteria with the ability to degrade vegetable packaging LDPE film were isolated from the waste disposal site located in Mathuradaspura, Jaipur, Rajasthan. Using the clear zone approach, the isolates were screened for LDPE biodegradation. In the presence of LDPE powder, isolate HSK30 was able to develop and produce a distinct clear zone around the colony. Weight loss analysis has been done after 120 days of incubation to assess the biodegrading capability of the isolate. Isolate HSK30 substantially reduced the weight of LDPE film (16.89 ± 1.02%) after 120 days as compared to control. Furthermore, efficacy of isolate in LDPE biodegradation was confirmed using biofilm formation, hydrophobicity, fourier transform infrared (FTIR) spectroscopy, scanning electron microscopy (SEM), and energy dispersive X-ray (EDX) analysis. Isolated strain HSK30 was identified as Exiguobacterium sp. using 16S rRNA gene sequencing and phylogenetic analysis. Thus, Exiguobacterium sp. strain HSK30 showed promising findings for vegetable packaging LDPE degradation and might be further investigated to improve its effectiveness in LDPE biodegradation. To the best of our knowledge, this is the first study to report the biodegradation of vegetable packaging LDPE film by Exiguobacterium sp.
