Atriplex halimus L. from two geo-climatic zones in North-Eastern Algeria: phenolic composition, anti-quorum-sensing, antibiofilm, anti-swarming, and enzyme inhibitory properties

Different solvent fractions were prepared from Atriplex halimus from two geo-climatic Regions of North-Eastern Algeria, Tebessa (T) and Negrine (N). Crude extraction yields were 21.00% and 21.34% for plant material from Tebessa (T) and Negrine (N), respectively, indicating that climate and altitude affected phytochemical content. HPLC–DAD indicated caffeic acid, p-coumaric acid, ferulic acid and rutin in significant amounts in both extracts, while quercetin was detected only in the plant from Tebessa. Fractions inhibited quorum-sensing mediated violacein production by C. violaceum CV12472 and C. violaceum CV026 at minimal inhibition concentration (MIC) and sub-MIC. Extracts showed concentration-dependent inhibition of swarming motility against flagellated P. aeruginosa PA01 and antimicrobial potentials against S. aureus, E. coli, and C. albicans. Fractions inhibited biofilm formation against S. aureus, E. coli, and C. albicans with S. aureus biofilm being the most susceptible to the fractions. The butanol fraction of plant from Negrine displayed highest anticholinesterase activity of 41.22 ± 0.86% (AChE) and 59.92 ± 0.75% (BChE) at 200 µg/mL. The ethyl acetate fraction of the plant from Negrine had highest antidiabetic activity, with an inhibition percentages of 56.18 ± 0.42% and 35.50 ± 0.38% against α-glucosidase and α-amylase, respectively. Results suggest that environmental conditions and altitude affect chemical composition and bioactivities of Atriplex halimus.
