Strain-resolved nutritional and bioactive composition of Indian-origin Lentinula edodes revealed through integrated analytical profiling

Reliable food composition data require strain-resolved measurements generated using analytically standardized methodologies; however, such reference data remain limited for Indian-origin Lentinula edodes strains, despite its growing importance in commercial cultivation and food applications. In this study, five commercially relevant Indian-origin L. edodes strains (DMRO-34, DMRO-35, DMRO-356, DMRO-623, and DMRO-388s) were systematically characterized using an integrated analytical profiling framework tailored for food composition assessment. Proximate composition, mineral elements, water- and fat-soluble vitamins, amino acid profiles, β-glucans, and selected bioactive-associated and metabolically relevant indices (total phenolics, total flavonoids, and free amino acids) were quantified using validated spectrometric, chromatographic, and enzymatic methods to ensure analytical comparability and reproducibility. Statistically significant strain dependent variation was observed across all compositional dimensions (p < 0.05). Protein (20.5–32.7% dry weight), β-glucans (15.4–30.4% dry weight), total phenolics (1.02–2.31 mg gallic acid equivalents g⁻1 dry weight), and free amino acids (0.094–0.147 mg g⁻1 dry weight) showed the greatest dynamic ranges. Multivariate analysis resolved clear strain level differentiation and revealed coordinated variation among nutritional and bioactive-associated traits. Within the evaluated dataset, strain DMRO-356 was positioned toward relatively higher values across multiple compositional parameters, including protein quality indices, essential minerals, β-glucans, and phenolic–flavonoid content. Comparison with published datasets indicates that the compositional ranges of Indian-origin strains fall within, and in several cases approach the upper bounds of, values reported for globally cultivated L. edodes. Overall, this study provides a strain-resolved, analytically robust compositional reference for Indian-origin L. edodes strains supporting improved food composition characterization, strain differentiation, and nutrition-informed evaluation.
