Characterization of polysaccharide conjugates extracted with alkali from tea residues—the byproducts of green tea beverage manufacturing

Effective utilization of tea residue is an important part of the comprehensive development of tea by-products. In this study, tea polysaccharide conjugates (TPC) were extracted from tea residues by 2% and 4% NaHCO3, named TPC-A2 and TPC-A4. TPC-A2 was composed of one homogeneous component with a molecular weight of 2.42 × 106 Da, and TPC-A4 was composed of two homogeneous components with molecular weights of 3.88 × 106 Da (96.24%) and 1.54 × 106 Da (3.76%). The protein portion of TPC-A2 contains 16 kinds of amino acids, while TPC-A4 contains 12 kinds. Possibly due to the shielding effect of the polysaccharide chains, their protein portion does not bind to the Coomassie Brilliant Blue dye G250. The zeta potential value was negative in both acidic and alkaline aqueous solutions, indicating that the sample was an acidic polysaccharide conjugate. The CD spectrum of TPC-A2 showed two positive Cotton effect peaks, while the CD spectrum of TPC-A4 showed no significant Cotton effect peaks. In addition, TPC-A2 and TPC-A4 also had the same strong hygroscopic ability as polysaccharide conjugates prepared from fresh tea leaves. These results will promote the exploitation and utilization of polysaccharides in tea residue.
