Medium modification of Cynanchum wilfordii adventitious root culture for the production of C21 steroidal glycosides, polysaccharides, and polypeptides using orthogonal design methods

Cynanchum wilfordii (Maxim.) Hemsl. is a perennial herbaceous plant containing various bioactive compounds, including C21 steroidal glycosides, polysaccharides, and polypeptides. Adventitious root (AR) culture is a promising approach for the production of C. wilfordii plant materials. In this study, single-factor and orthogonal design experiments were conducted to modify the Murashige and Skoog (MS) basal medium for C. wilfordii AR culture; the adjusted factors included nitrogen, ammonium to nitrate (NH4+/NO3−) ratio, phosphorus, and calcium concentrations. Results showed that optimal AR biomass and contents of total C21 steroidal glycosides, polysaccharides, and polypeptides were obtained at four different combination groups of nitrogen, NH4+/NO3−, phosphorus, and calcium concentrations. In comparison, maximum productivities of total C21 steroidal glycosides, polysaccharides, and polypeptides were observed in the combination of 30.0 mM nitrogen with 1/2 NH4+/NO3−, 1.5 mM phosphorus, and 1.5 mM calcium. After medium modification, productivities of C21 steroidal glycosides (approximately 1.9 g L−1) was 4.7-fold, polysaccharides (approximately 2.8 g L−1), and polypeptides (approximately 1.4 g L−1) were more than threefold higher than in standard MS medium. The finding of this study provided a reference for mass production of C. wilfordii ARs enriched bioactive compounds.
