Effect of BA and TDZ on micropropagation of Myosotis sylvatica Hoff. (forget-me-not)

Myosotis sylvatica Hoff. is one of the important perennial, herbaceous species used as a ground cover ornamental plant in gardens. This plant is also utilized in various industries, including the pharmaceutical and cosmetic sectors. Because of these properties, mass propagation of M. sylvatica is significant. Tissue culture provides many advantages for both mass production and in vitro conservation of plants. In this study, M. sylvatica seeds germinated on a plant growth regulator-free (PGR-free) Murashige and Skoog (MS) medium. Shoots derived from seedlings were cultured on MS medium including 30.0 g L−1 sucrose, 4.0 g L−1 gelrite, 0.1 mg L−1 1-naphthaleneacetic acid (NAA), and different concentrations of benzyladenine (BA) and thidiazuron (TDZ) (0.3, 0.5, 1.0, 2.0, and 3.0 mg L−1). Shoot formation, multiplication rate, and rooting of the explants were observed. BA was more effective than TDZ for shoot regeneration according to the t-test. Efficient multiplication was obtained from MS medium including 0.1 mg L−1 NAA and 2.0 mg L−1 BA. The highest multiplication rate was observed from MS medium, including 0.1 mg L−1 NAA and 2.0 mg L−1 BA as 2.04 ± 0.5 and 3.86 ± 0.51 shoot per explant in the first and second subcultures, respectively. Shoot explants were cultured on MS medium, including different NAA (0, 0.1, 0.3, and 0.5 mg L−1) concentrations for root initiation, and the highest root per explant formed in MS medium supplemented with 0.1 mg L−1 NAA. The highest root length was measured as 0.42 cm on MS medium, including 0.1 mg L−1 NAA. Rooted plants were acclimatized, and the survival of the plant was evaluated for 4 wk. This is the preliminary study for the in vitro propagation of M. sylvatica, and a micropropagation protocol was developed for this ornamental ground cover plant.
