pH characteristics and calcium carbonate dissolution potential of Aspergillus section Restricti isolated from house dust

The effect of pH on mycelial growth and ability to dissolve CaCO3 were evaluated for 101 strains from 17 Aspergillus section Restricti species isolated from house dust in Japan. The studied strains evaluated the (1) mycelial growth on dichloran 18% glycerol or potato dextrose agar + 15% NaCl adjusted to pH 3, 5, 7, 9, and 11 and (2) ability to dissolve CaCO3 on CaCO3 glucose agar supplemented with 22% glycerol or 15% NaCl to adjust the water activity. Four series Restricti species (A. conicus, A. domesticus, A. gracilis, and A. pseudogracilis) showed optimal growth at a weakly acidic pH (pH 5) compared to other series. Two series Restricti species (A. caesiellus and A. restrictus) and all series Penicillioides species showed greater mycelial growth at pH 11 than the other series. Three series, excluding series Halophilici, dissolved CaCO3. CaCO3 dissolution abilities of six series Restricti species (A. conicus, A. domesticus, A. gracilis, A. pseudogracilis, A. restrictus, and A. villosus) and A. vitricola (series Vitricolarum) were stronger than those of species in other series. These results indicate that the species with different physiological characteristics are distributed in house dust. The species that have been detected in marine and limestone substrates by previous studies were found to have pH characteristics and CaCO3 dissolution abilities suitable for survival in their respective environments.
