Purpose <p>In this study, we analyzed the effects of moderate-intensity interval training on the nod-like receptor (NLR) family pyrin domain-containing protein-3 (NLRP3) inflammasome pathway in the liver tissue of male rats with type 2 diabetes. </p> Methods <p>In this experimental study, 20 male Wistar rats were arbitrarily assigned to 4 groups: (1) a healthy control group (Con), (2) a group with moderate-intensity interval training (MIIT), (3) diabetes group (Di), and (4) diabetes + moderate-intensity interval training group (Di + MIIT). Diabetes was induced by a combination of high-fat diet (HFD) and streptozotocin (STZ). MIIT was scheduled for 8 weeks, with sessions occurring four times per week on a treadmill at an intensity of 60–70% of maximum oxygen uptake (VO2max). Forty-eight hours after training, the liver tissue was removed under sterile conditions, and the NLRP3 and ASC protein expression was identified using the western blot method. Interleukin (IL)-1β and IL-18 concentrations were estimated in tissue lysate utilizing a species-specific ELISA kit as recommended by the producer’s guideline. We also measured lipid profile (low-density lipoprotein (LDL), high-density lipoprotein (HDL), cholesterol, as well as triglyceride (TG)) from blood samples of rats. One-way ANOVA and Tukey post hoc test were utilized to compare means.</p> Results <p>The induction of diabetes significantly enhanced the expression of NLRP3 proteins, apoptosis-associated speck-like protein containing a CARD (ASC), tissue levels of IL-1β, IL-18, and serum levels of TG, LDL, while reducing HDL levels (<i>p</i> &lt; 0.05). The Di + MIIT group showed reduced NLRP3 expression, ASC, tissue levels of IL-1β, IL-18, and serum levels of TG, LDL, and cholesterol, except HDL. The difference between NLRP3 protein expression, tissue levels of IL-1β, IL-18, and serum lipid profile between the MIIT and control groups was not significant (<i>p</i> &gt; 0.05); in contrast, the MIIT group increased ASC protein expression in the liver tissue (<i>p</i> &lt; 0.05).</p> Conclusion <p>These findings highlight the therapeutic potential of moderate-intensity interval training as a non-pharmacological intervention to manage liver inflammation associated with type 2 diabetes, warranting further investigation into its mechanisms and long-term benefits.</p> Graphical Abstract <p></p>
