Acute moderate exercise reduces the serum SPARC levels in older adults: effect modification by blood PYCARD methylation

Secreted protein acidic and rich in cysteine (SPARC) is a key pro-inflammatory factor in aging-associated systemic inflammation. A few studies have investigated the acute effect of high-intensity exercise on circulating SPARC in younger subjects, but to date, there have been no such studies on moderate-intensity exercise in older individuals.
AimsThe purpose of the current study was to clarify the acute response of moderate bench-step exercise on serum SPARC levels in older adults. We also examined whether the individual alterations in SPARC levels after the bench-step exercise might be influenced by the blood DNA methylation levels of PYD and CARD domain-containing protein (PYCARD) gene, another key inflammatory factor, rather than SPARC.
MethodsThirty-six older adults (7 men and 29 women) performed a single bout of moderate bench-step exercise at their anaerobic threshold for 15 min. Serum SPARC concentrations were measured before and immediately after the exercise. Pre-exercise PYCARD methylation levels in the peripheral blood were also measured.
ResultsThe serum SPARC was significantly decreased by 30% immediately after the 15-min bench-step exercise (pre-exercise: 531 ± 132 ng mL−1; post-exercise: 372 ± 138 ng mL−1; P < 0.01). Higher PYCARD methylation (at CpG site 3) was associated with a greater decrease in serum SPARC levels immediately after bench-step exercise (r = − 0.35, P < 0.035).
ConclusionsAn acute bout of moderate bench-step exercise at the anaerobic threshold reduces serum SPARC levels in older adults. The individual differences in the beneficial effect of acute moderate exercise on SPARC may be modified by pre-exercise blood PYCARD methylation status.
