Association of single nucleotide polymorphisms in the entire coding region of the SOCS2 gene with milk traits in Sahiwal cattle

This study investigated polymorphisms in the entire coding region of Suppressor of Cytokine Signalling 2 (SOCS2) gene and their association with milk traits in Sahiwal cattle: First lactation 305 days milk yield (Kg) (FL305MY), Average test day milk yield (Kg) (ATDMY), Average test day fat percentage (%) (ATDFP) and Lactational fat yield (Kg) (LFY). The traits ATDMY and ATDFP were corrected for significant non-genetic factors by a fixed linear model and subsequently used in association studies. The Animal model was used to estimate FL305MY and LFY breeding values. The average FL305MY, ATDMY, ATDFP and LFY were estimated as 1816.56 ± 40.79 kg, 6.61 ± 0.11 kg, 4.76 ± 0.02% and 84.68 ± 2.00 kg, respectively. Six sets of primers exhibited a total of 05 SNPs including 04 transitions and 01 transversion in the entire coding region. Association analysis revealed significant (p < 0.05) effect of SNPs T219C and A280G on LFY whereas, other SNPs had no significant effects on milk traits. The GGAA haplotype was strongly (p < 0.001) linked to an increase in LFY. Allele substitution effects at T219C and A280G further validated the findings, with increasing LFY by 2.687 ± 0.8% and 1.493 ± 0.797%, respectively. The GGAA haplotype increased LFY by 1.531 ± 0.187%. The SNPs T219C and A280G, along with GGAA haplotype, are potential genetic markers for marker-assisted selection aimed at enhancing milk fat production.
