<p>This study assessed preovulatory follicular development in ewes and, consequently, the synchronization of ovulations induced by C6 (Compound C6), a synthetic kisspeptin (Kp) analogue with an extended half-life. The estrus of 30 anestrous Corriedale ewes was synchronized using intravaginal sponges (IVS) containing medroxyprogesterone acetate for 14 days. The experimental groups were: (1) GeCG received 300 IU eCG at IVS withdrawal; (2) GC6 received 15 nmol of C6 24&#xa0;h after the removal of the IVS; and (3) Gcon, which was only treated with the IVP. GeCG and Gcon exhibited a higher number of ewes in estrus compared to GC6 (10/10, 7/10, 2/10; <i>P</i> &lt; 0.0001). The time from IVS withdrawal to ovulation did not differ among groups (Gcon: 57.0 ± 2.8&#xa0;h; GeCG: 60.1 ± 2.7&#xa0;h; GC6: 52.1 ± 2.8&#xa0;h; <i>P</i> = 0.14). Ovulations were more synchronous in GC6 than in GeCG and Gcon (SD: 4.1, 10.7, and 8.3&#xa0;h, respectively, <i>P</i> = 0.016). The frequency of ewes with more than one CL was higher in GC6 (7/10) and GeCG (7/9) compared to Gcon (2/10) (<i>P</i> = 0.002). The diameter of ovulatory follicles was greater (<i>P</i> = 0.02) in Gcon ewes (7.1 ± 0.8&#xa0;mm) than in GeCG ewes (6.4 ± 0.2&#xa0;mm), but GC6 (7.0 ± 0.6&#xa0;mm) did not differ from either group. No differences were observed in the quality of the CL. In conclusion, based on these results, C6 is a potential alternative to eCG in ovulation synchronization protocols for ewes, as it stimulates similar follicular development and more synchronized ovulations. Deeper studies should be performed considering hormonal measurements and flock fertility.</p>
