<p>Bovine leukemia virus (BLV), in addition to causing enzootic bovine leukosis (EBL), has been suspected as the cause of breast cancer in humans in the last decade. Since milk is one of the ways of excretion of the virus and transferring it to other cows or humans, this study was conducted to investigate the correlation between infection and shedding provirus BLV through milk. For this purpose, blood and milk samples were taken from 160 cows from 4 dairy farms in Tehran and Isfahan provinces of Iran. To detect antibodies to BLV, sera were tested by a commercial ELISA kit and Nested PCR were used for detection of BLV provirus in milks. Overall, 82.9% of the studied cows had antibodies to BLV in sera and 31.87% of them shed BLV provirus from milk. According to the result of ELISA and PCR, there was a significant difference between farms of Tehran and Isfahan. In addition, statistical analysis showed no-correlation between ELISA and milk PCR in all cows (Kappa = 0.038, <i>P</i> &lt; 0.001). It is concluded that, although there is no significant correlation between infection with the virus and its shedding through milk, 31.9.2% of infected cattle shed the virus through milk, and there is a risk of infecting other cows and humans in this way. If it is proven that BLV is the definitive cause of breast cancer in humans, how to prevent its transmission through milk such as pasteurization should be investigated.</p>
