Meta-topolin increases the number of shoots and rhizogenesis during in vitro propagation of a Laeliocattleya hybrid

The micropropagation of orchids is a well-established strategy in plant biotechnology, primarily driven by their high ornamental and commercial value. To optimize propagation protocols, this study evaluated the effect of meta-topolin (mT) on the micropropagation of Laeliocattleya canhamiana, a high-value ornamental hybrid. Shoot tips were cultured on Murashige and Skoog (MS) medium supplemented with 30 g L⁻¹ sucrose and 1 mg L⁻¹ naphthaleneacetic acid (NAA), while different concentrations of mT (0, 0.5, 1.0, and 1.5 mg L⁻¹) were tested across two 30-day subcultures. After 30 days, shoot number, shoot length, leaf number, root number, and root length were recorded. Acclimatization was performed in a sterile substrate composed of peat and agrolite, and survival rates were evaluated after 180 days. The greatest shoot proliferation was obtained at 1.5 mg L⁻¹ mT, followed by 1.0 mg L⁻¹, which also enhanced shoot elongation, root number, and root length. Following acclimatization, survival rates reached 95%. These findings provide a reproducible and efficient protocol that strengthens the commercial micropropagation potential of this valuable orchid hybrid.
