<p>Hairy root (HR) cultures represent a valuable biotechnological platform for the production of bioactive compounds. In this study, an optimized protocol was developed for the induction and maintenance of HR cultures from the Argentine peanut (<i>Arachis hypogaea</i> L.) cultivar Granoleico, using <i>Rhizobium rhizogenes</i> strain LBA9402. Cotyledon explants exhibited higher transformation efficiency (80%) when cultivated in solid Hoagland (Hg) medium compared to the conventional half-strength Murashige and Skoog (½ MS) medium. Among the independent HR lines obtained, line C9 showed the highest growth index (GI = 21.8) and growth rate (GR = 165.9 mg day<sup>−1</sup>) in liquid Hoagland medium supplemented with vitamins (HgVit). Biochemical characterization of C9 revealed that unsaturated fatty acids accounted for 71% of total lipids, with linoleic (36.5%) and linolenic acids (23.3%) as the most abundant. The total phenolic content reached 2982.7 mg GAE g<sup>−1</sup> FW, and antioxidant activity was 2.15 mM AAE g<sup>−1</sup> FW (24% DPPH inhibition). HPLC-MS analysis identified four flavonoids, predominantly naringin (1.62 ng mg<sup>−1</sup> FW) and genistein (0.45 ng mg<sup>−1</sup> FW). These findings demonstrate that peanut HR cultures from cv. Granoleico constitutes an efficient in vitro system for the production of antioxidant- and lipid-rich metabolites with potential applications in food, pharmaceutical, and agricultural biotechnology.</p>
