Agrobacterium-mediated transformation for gene editing in tomato: challenges and advances

Agrobacterium-mediated transformation remains the most effective and widely used method for introducing foreign genes into tomato, a model crop among dicotyledonous species for developmental and functional studies. Despite the availability of standardized protocols for model tomato cultivars, transforming recalcitrant non-model tomato cultivars remains as a challenge due to genotype-dependent responses to tissue culture conditions. Over the past decades, tomato transformation has undergone substantial refinement with growing emphasis on cultivar-specific optimization strategies. Furthermore, more research and efforts toward developing versatile, genotype-independent tomato transformation protocols are essential for expanding and fully utilizing the benefits of evolving genome editing tools for crop improvement in tomato. This mini-review provides insights into critical factors affecting successful tomato regeneration and transformation, and also highlights some of the strategies deployed in recently optimized protocols for overcoming these challenges.
