Development of an in vitro propagation system for ornamental Calathea fasciata ‘Borrusica’

Given the commercial importance and propagation difficulties of Calathea fasciata ‘Borrusica’ in horticultural production, this study systematically optimized the in vitro rapid propagation techniques for this ornamental foliage plant. Various factors affecting propagation efficiency were evaluated, including disinfection methods for explants, selection of direct or indirect regenerated explants, and the effects of different types or concentrations of plant growth regulators (PGRs) on shoot proliferation and root formation. Despite the challenges associated with explant sterilization, this study represents the first report on the feasibility of using Bijieshi solution (BJS) for explant disinfection in Calathea. Among 4 different types of explants, basal shoot fragment was identified as the most optimal explant for shoot induction. MS medium supplemented with 2.0 mg L⁻¹ 6-Benzylaminopurine (BA) and 0.2 mg L⁻¹ α-naphthalene acetic acid (NAA) was optimized for shoot induction, achieving an induction rate of 93.98%. The 95.56% rooting rate was achieved on 1/2MS medium supplemented with 0.5 mg L⁻¹ NAA and 96.00% survival rate following acclimatization. This study provides significant foundation for advancing commercial cultivation of C. fasciata ‘Borrusica’.
