Improved culture conditions for successful micropropagation of smoke tree Cotinus coggygria ‘Royal Purple’ through direct organogenesis

Smoke tree Cotinus coggygria has not only high ornamental value but also important medicinal properties. The present research studied the effect of some factors on aseptic cultures establishment, in vitro shoot multiplication, in vitro rooting and ex vitro acclimatization of the economically important cultivar C. coggygria ‘Royal Purple’. In order to establish the aseptic cultures of C. coggygria ‘Royal Purple’, axillary shoots from dormant twigs dipped in water in February and March were sterilized with 70% (v/v) ethanol for 20 s and with 0.1% (w/v) HgCl2 solution for 6 min, soaked in 150 mg L−1 vitamin C solution for 10 min and then inoculated on the shoot induction medium which was Woody Plant Medium (WPM) supplemented with 1.0 mg L−1 6-benzyladenine (6-BA), 0.05 mg L−1 1-naphthaleneacetic acid (NAA), 1000 mg L−1 polyvinylpyrrolidone (PVP), 3% sucrose and 0.6% agar, resulting in the browning-necrosis rate of 2.5% and the in vitro shoot induction of 79.5%. In vitro shoots were multiplied on modified Murashige and Skoog (MS) medium (NH4NO3 1100 mg L−1) enriched with 1.0 mg L−1 6-BA, 0.05 mg L−1 NAA, 1000 mg L−1 PVP, 3% sucrose and 0.6% agar, resulting in the shoot proliferation rate of 4.6 fold without hyperhydricity. In vitro rooting was achieved in 1/2 MS medium supplemented with 1.5 mg L−1 indole-3-butyric acid (IBA), 1000 mg L−1 PVP, 2% sucrose and 0.6% agar, with a root induction rate of 86.8%. The suitable pre-hardening period for greenhouse acclimatization of in vitro rooted plantlets was about 3–5 days, the early humidity-hardening period was 11–13 days, the appropriate in vitro plantlet height for rooting induciton was ~ 4.0 cm and the best acclimatization medium was a mixture of vermiculite, peat, carbonized rice husk, fermented sawdust and coal ash (at a volume ratio of 2:2:4:1:1). Under these conditions, an acclimatization rate of about 80% was achieved after 35 days. The rooted and acclimatized plants were successfully transferred to soil after light hardening under sunlight for 7–9 days resulting in 95% survival.
