Administrating effectual direct regeneration of microtubers and shoots from various in vitro root types of Cyclamen persicum Mill.

Cyclamen persicum Mill. is recognized as one of the most important potted plants globally. Here, we investigated the microtuber and shoot induction directly from different types of in vitro roots as a novel and time-reserving method for direct organ regeneration from hybrid cultivars of C. persicum Mill. After optimizing the best hormonal composition for regeneration, the effect of the root types (seedling, adventitious, and secondary) and plate sizes (6, 8, and 10 cm diameter Petri dishes) were evaluated in different experiments. The best hormonal treatment for direct regeneration of microtubers and shoots was 1.0 mg·L−1 benzyladenine and 0.1 mg·L−1 α-naphthaleneacetic acid for Cyclamen cv. Pure White after 8 weeks incubation at 24 ± 1 °C in darkness. Accordingly, the maximum number of shoots per explant (3.87) and the maximum number of microtubers per explant (5.58) were recorded for Cyclamen cv. Pure White in a Petri dish size of 10 cm diameter. The latter values were 5.3 and 3.5 for Cyclamen cv. Neon Pink and 2.5 and 2.1 for Cyclamen cv. Dark Violet, respectively. The highest weight of regenerated microtubers (3.4 g) was obtained from in vitro adventitious roots of Cyclamen cv. Pure White cultured on a Petri dish size of 10 cm. However, there was no significant difference between regeneration percentages depending on the types of in vitro roots. The ability to use different types of in vitro roots for direct shoot and microtuber regeneration can be employed to develop a convenient protocol for the commercial micropropagation of C. persicum Mill. as well as the transformation approaches.
