Transcriptome-wide identification of MYB gene family and functional analysis of LcsMYB123 in Litsea coreana var. sinensis

Litsea coreana Levl. var. sinensis (Allen) Yang et P. H. Huang serves as the predominant botanical source for Hawk tea, known for its high flavonoid content, which contribute to its health-promoting benefits in humans. MYB transcription factors (TFs) are one of the largest gene families in plants and play a central role in regulating flavonoid biosynthesis. However, the MYB gene family in L. coreana var. sinensis has not been comprehensively identified and characterized. In this research, a transcriptome-wide analysis of MYB TFs was performed and 150 MYB genes were identified in L. coreana var. sinensis, including 31 1R-MYB, 113 R2R3-MYB, 5 3R-MYB, and 1 4R-MYB. Phylogenetic analysis indicated that 113 R2R3-MYB TFs can be divided into 32 subgroups. Here, the function of an R2R3-MYB TF, LcsMYB123, which grouped with AtMYB123, was further verified. Subcellular localization experiments showed that LcsMYB123 was localized in the nucleus and cytoplasm. Overexpression of LcsMYB123 in Arabidopsis resulted in higher flavonoid content, indicating that LcsMYB123 functions as a positive regulator of flavonoid biosynthesis. These findings establish a foundation for the study of MYB TFs in L. coreana var. sinensis and contribute significant insights for subsequent research into the potential roles of MYB genes in flavonoid biosynthesis.
