Study on physiological and biochemical characteristics being used for identification of different plantlet regeneration approaches from somatic embryo of Rosa hybrida ‘J. F. Kennedy’

During somatic embryo culture of Rosa hybrida ‘J. F. Kennedy’, there were two regenerated materials with distinct morphological differences, one was a regenerated-seedling-with-a-single-bud-along-the-longitudinal-axis (RSLA seedling), the other were regenerated-multiple-buds-along-the-horizontal-axis (RMBHA buds). Through the measurement and analysis of physiological and biochemical indicators of these cultured embryogenic materials, it showed that the regeneration process of rose somatic embryos could be distinguished between the initial and regenerated materials, by comparing the activities of protective enzymes which the indicators included SOD, POD and CAT activities, and the levels of membrane lipid peroxidation products which the indicator was MDA content. Additionally, during the development processes of the two types of regenerated buds, the activities of SOD and POD were significantly lower in that of RSLA seedling than in that of RMBHA buds. This study identified the regeneration processes of different plant materials based on these characteristic changes in these above indices, and the disparities in vital activities might provide insights for molecular analysis of the regeneration process for rose ‘J. F. Kennedy’.
