Influence of the nitrosyl iron complex with thiosulfate ligands on carbohydrate metabolism and lipid peroxidation parameters in mice with experimental diabetes mellitus

Tetranitrosyl iron complex (TNIC) has pronounced antioxidant and antiglycation properties and is also an inhibitor of aldose reductase. Its in vivo influence on models of type 2 diabetes mellitus (T2DM) induced by a combination of high-fat diet and low-dose injections of streptozotocin in C57Bl/6J mice was studied. The daily administration of TNIC for 14 days to mice with T2DM at doses of 9.4 and 15.7 mg kg−1 had a pronounced antihyperglycemic effect: the glucose level in the blood decreased appreciably to the 12th week of the experiment. The TNIC in a dose of 15.7 mg kg−1 exerts a stronger antihyperglycemic effect compared to a dose of 9.4 mg kg−1 and the classical NO-donor diethylenetriamine NONOate decreasing the glucose level nearly to the level of healthy mice. In addition, the animals demonstrated a significant decrease in the level of lipid peroxidation in the blood plasma and liver. The obtained results confirm that the TNIC has a high potential for use in the therapy of T2DM and its complications.
