Research progress on chemistry, bioactivity, and NMR patterns of acetylene compounds in natural products (2015–2025)

Acetylenes, a class of bioactive small-molecule natural products featuring carbon–carbon triple bonds, are predominantly distributed in higher plant families such as Asteraceae, Apiaceae, Araliaceae, and Campanulaceae. whereas they occur relatively rarely in animals and microorganisms. Their broad pharmacological potential has attracted significant interest, yet a systematic analysis of their nuclear magnetic resonance (NMR) characteristics remains lacking. This review comprehensively covers 296 natural acetylenes (258 linear and 38 cyclic) reported between 2015 and 2025. Particular emphasis is placed on the detailed NMR spectral patterns and structural diversity of these compounds, alongside their wide-ranging biological activities, including antitumor, anti-inflammatory, neuroprotective, lipid metabolism-regulating, and antimicrobial properties. This review elucidates the structural specificity imparted by the carbon–carbon triple bond and provides a systematic analysis of the corresponding NMR characteristics and biological activities, thereby laying a valuable foundation for the structural identification and future development of acetylene-containing natural products.
