Simultaneous Determination of Paracetamol and Chlorzoxazone by Derivative Spectrophotometric and Chromatographic Methods

A second-derivative spectrophotometric method was developed to analyze paracetamol (PAR) and chlorzoxazone (CZN) simultaneously. The calibration wavelengths were determined at the zero crossing points of the second-order derivative spectra, and the coefficients of determination (R2) were 1.00 at 278.5 and 290.3 nm for PAR and CZN. The method’s limit of detection (LOD) values were 0.24 mg/L and 0.20 mg/L, and the limit of quantification (LOQs) values were 0.80 mg/L and 0.67 mg/L for PAR and CZN. The calculated recoveries from the synthetic mixture were 100.4% for PAR and 98.2% for CZN, and the results were compared with the HPLC method. Methanol was used as the mobile phase, and a C18 column was used as the stationary phase in the analysis. The absorption of PAR and CZN was measured at 260 nm, and the calculated coefficient of determination (R2) values were 0.9999 and 0.9994. The same synthetic mixtures were used for recovery studies, and 99.2% and 99.8% values were calculated for PAR and CZN.
