<p>This research endeavors to explore the bioactive potential of <i>Ehretia laevis</i> Roxb. roots, sourced from Burkoni, Tal. Hinganghat, DistrictWardha, Maharashtra, India. The study employs a systematic approach, utilizing Soxhlet extraction to obtain the crude ethanol extract from the roots. Subsequent fractionation is carried out employing solvents of varying polarity, leading to the isolation of distinct fractions. High-performance thin-layer chromatography fingerprinting is employed to analyze the obtained fractions. Acute toxicity studies are conducted to assess the safety profile of the crude extract. The antimicrobial activity of the crude extract and its fractions is investigated, showcasing promising results. Preliminary characterization of various fractions provides insights into their composition. Furthermore, bioactivity-guided isolation using column chromatography results in the effective separation of five distinct compounds, labeled NH-I, NH-II, NH-III, NH-IV, and NH-V. Minimal inhibitory concentration (MIC) determination is carried out to evaluate the potency of these isolated compounds against microbial pathogens. The antimicrobial activity of the isolated compounds is scrutinized, emphasizing their potential as novel antimicrobial agents. Structural elucidation of NH-I is accomplished through advanced spectroscopic techniques, including <sup>1</sup>H NMR, <sup>13</sup>C NMR, and mass spectroscopy, shedding light on the molecular composition and configuration of this bioactive compound. In conclusion, this research unfolds a comprehensive exploration of the antimicrobial potential of <i>Ehretia laevis</i> Roxb. roots, offering valuable insights into the bioactive compounds present in the plant and their potential applications in medicine and pharmacology.</p>
