Antiparkinsonian Effect of Ladasten on a 6-Hydroxydopamine-Induced Parkinsonian Syndrome Model in Mice

The study focused on the antiparkinsonian effect of ladasten (10, 50, 100 mg/kg) on a parkinsonian syndrome model in mice induced by unilateral administration of 6-hydroxydopamine. The ability of ladasten (most effectively at doses of 50 and 100 mg/kg) to reduce the severity of various manifestations of parkinsonian syndrome in rotarod tests at fixed and accelerating rotation speed, the pole test, the cylinder test, and the footprint test as compared with an active control was established. The ability of ladasten to exert a stimulating effect on the synthesis and metabolism of dopamine in the used model was also established and was most effective at a dose of 100 mg/kg because the dopamine level was 3.8 (p < 0.001); 3,4-dihydroxyphenylacetic acid, 2.2 (p < 0.01); and homovanillic acid, 2.1 (p < 0.01) times higher than that of the active control.
