<p><i>Zygophyllum cornutum</i> Coss. is used for treatments of various ailments such as inflammatory, rheumatism and diabetes. This work aims to evaluate the impact of extraction with different polarity solvents on the phytochemical content and therapeutic ability of <i>Zygophyllum cornutum</i> Coss. The phenolic profile of the infusion, <i>n</i>-butanol and ethereal extracts was established using high-performance liquid chromatography (HPLC) and total phenolic contents. The pharmacological properties were performed <i>in vivo</i> and <i>in vitro</i> using analgesic, anti-inflammatory, antioxidant assays and their acute toxicity test. The results showed a positive correlation between the polarity of solvent, phytochemical content and pharmacological properties. The phytochemical analysis revealed that the infusion extract presents the maximum number of compounds identified compared to other extracts and a higher amount of total phenolic compounds (71.12 ± 0.20 mg EAG/g extract) followed by <i>n</i>-butanol extract (45.18 ± 0.11 mg EAG/g extract) and then ethereal extract (24.00 ± 0.21 mg EAG/g extract). The infusion extract is the more effective, followed by the extracts of <i>n</i>-butanol, and then ethereal. Indeed, the infusion extract possesses high antioxidant, analgesic and anti-inflammatory effects superior toor equal to the reference drugs. Acute toxicity test showed that the three <i>Zygophyllum cornutum</i> Coss. extracts are innocuous. The present study indicates that the different solvents and extraction methods can affect the phytochemical content and pharmacological properties of <i>Zygophyllum cornutum</i> Coss. Water is the optimal solvent for the extraction of active compounds from <i>Zygophyllum cornutum</i> Coss. aerial parts, notably quercetin and salicylic and ascorbic acids, which can be responsible for strong antioxidant, analgesic, anti-inflammatory activities.</p>
