HPLC Determination of Rutin in the Aerial Part of Glycyrrhiza glabra L.

Alcohol extracts from the herb of Glycyrrhiza glabra L. were studied by HPLC on a Supelco Analytical C18 chromatographic column (4.6 mm × 15 cm, particle size 5 μm) eluted with a mobile phase of 0.1% formic acid and MeCN (50:50, v/v), column temperature 30°C, and extinction at 205 nm. The rutin peak, the retention time of which coincided with that of its reference standard used in the study, clearly dominated the chromatograms. The content of biologically active rutin in the aerial part of the medicinal plant ranged from 2.59 to 2.79%. The error of a single determination of rutin in licorice herb was ±3.08% with 95% confidence probability. The obtained data substantiated the choice of rutin reference standard for use in the quantitative determination of flavonoids in the aerial part of Glycyrrhiza glabra by differential spectrophotometry.
