Mechanism of Myogenesis Stimulation by Ethylmethylhydroxypyridine Succinate

The search for new targets for pharmacological stimulation of myogenesis and muscle regeneration is an urgent issue. The aim of the study was to reveal the role of succinate receptors (SUCNR1) in stimulating myogenesis under the action of ethylmethylhydroxypyridine succinate (EMHPS). The study was conducted on C2C12 cell line. Myogenic differentiation was stimulated by growth medium containing 2% horse serum. The effect of EMHPS on differentiation was evaluated by adding it to differentiation medium at concentrations of 1 – 1000 μM. Pertussis toxin inhibitor Gi (100 ng/mL) together with EMHPS was used to study the role of SUCNR1 in this process. The myogenesis index and the length and width of multinuclear myoblasts were evaluated on days 1, 4, and 7 of differentiation. The levels of myosin, myogenic determination factor (MyoD), myogenin (MyoG), and SUCNR1 were evaluated using Western blot. EMHPS was shown dose-dependently to accelerate myogenic differentiation of C2C12 cell line. This effect was realized through the SUCNR1-Gi signaling pathway which, in turn, could be considered a promising target for pharmacological actuation of the myogenic differentiation process.
