Age-related changes of leptin reception in the dorsomedial nucleus of the hypothalamus in rats

The sensitivity of the hypothalamus to the action of regulatory factors including hormones changes with aging. Leptin is one of the most important hormones regulating metabolism. Using immunohistochemistry and electrophysiological methods, we studied the expression of leptin receptors (LepR), as well as the effect of leptin on the firing rate of neurons in the dorsomedial nucleus (DMN) of the hypothalamus in male rats of different age groups (2–3 months, 12 months, and 24 months). The percentage of neurons in the hypothalamic DMN immunoreactive to LepR varied from 34.8 ± 5.3% to 38.1 ± 8.6% and did not change with age. The firing rate of DMN neurons in response to leptin administration also did not change significantly in different age groups. However, a change in the firing rate of most individual hypothalamic neurons was observed after leptin administration—from primarily inhibitory in the young and adult rats to predominantly activating in the aged rats. Thus, despite the constancy of the receptor composition, aging results in a qualitative change in the response of DMN neurons to the leptin.
