Relationship Between the BDNF Level in Areas of the Medial Prefrontal Cortex of Rats during Motivation-Driven Learning of a Formed Behavior: Approach/Avoidance

This article presents the results of a comparison of the extent of changes in molecular markers of systemogenesis during actualization of experience domains of approach and avoidance differing in terms of the level of involvement of stress-realizing mechanisms. Analysis of the content of brain-derived neurotrophic factor (BDNF) in the medial zones of the prefrontal cortex identified differences due to the motivation of the formed behavior: approach or avoidance. Studies were carried out on adult Long Evans rats. BDNF was detected by the indirect immunoperoxidase method on cryogenic brain sections. Preliminary behavioral phenotyping was used to form two groups learning a behavior consisting of pressing a pedal: 1) to start the feeder and get food; 2) to turn off the current and avoid pain. Quantitative analysis of immunoreactivity in the prelimbic and infralimbic zones of the prefrontal cortex revealed a significantly smaller number of BDNF-immunopositive cells in animals of the avoidance behavior training group compared to the approach behavior training group. An association between BDNF levels and individual characteristics such as anxiety and exploratory activity was also found. These studies led to the conclusion that the medial prefrontal cortex is involved in ensuring approach behavior, and also avoidance behavior, albeit to a significantly lesser extent. The probable role of stress-induced neuroinflammation in the differences found in these studies is discussed.
