Objectives <p>Hepatitis C virus (HCV) is a major global health burden. While, host genetic context was considered to influence its infection risk. Here we aimed to identify if polymorphisms in telomerase reverse transcriptase (<i>TERT</i>), a key player in aging including immune-senescence, belong to the context.</p> Methods <p>The study included a total 1166 case-control cohort with 502 Chinese HCV patients and 664 sex/age-matched healthy controls. Quantitative PCR was used to test <i>TERT</i> rs2736098 and rs2736100 genotyping and Leukocyte telomere length (LTL).</p> Results <p>The rs2736100_AC/CC genotypes were significantly associated with lower risk of HCV infection compared to the AA genotype (OR 0.770, 95%CI 0.603–0.983, <i>P</i> = 0.036). Further analyses showed that association between the telomerase rs2736100_CC genotype and a lower risk of chronic hepatitis C in Chinese males (OR 0.579, 95%CI 0.362–0.924, <i>P</i> = 0.022). There was no difference in the rs2736098 variants between controls and HCV patients. LTL and age-adjusted LTL were shorter in HCV patients compared with healthy adults, shorter telomeres with susceptibility to chronic hepatitis C.</p> Conclusions <p>The <i>TERT</i> rs2736100_CC genotype is associated with a lower HCV risk in Chinese males. LTL is shorter in HCV patients compared with healthy adults, which indicates that HCV has affected telomere length. These findings may have implications in HCV infector pathogenesis and prevention.</p>

Objectives
