Dietary Sargassum zhangii increased growth, hemato-immune responses and disease resistance against a fungal disease (Aphanomyces invadans) in Labeo rohita fingerlings

A 10-week study was conducted to evaluate the effects of raw or autoclaved Sargassum zhangii on rohu (Labeo rohita, 11.6 ± 0.14 g) fingerlings. Four experimental diets were supplemented with 2.5% (RSZ2.5) or 5% (RSZ5) of raw or 2.5% (ASZ2.5) or 5% (ASZ5) autoclaved S. zhangri meal. An algae-free diet served as a control diet (crude protein: 35.14%; crude Lipid: 10.01%). Fifteen 180-L rectangular reinforced plastic containers were stocked with 375 fingerlings (25 fish per container), which were split up into five experimental treatments, each in triplicate. Following the feeding trial, the fish received an intramuscular injection of the virulent fungal pathogen Aphanomyces invadans at a concentration of 104 spores mL−1, then their cumulative mortality was monitored for 30 days. Fish fed S. zhangii meal supplemented diets, particularly RSZ5 group had higher growth performance and feed intake compared to the control (P < 0.05). Supplementing diet with 5% raw or autoclaved S. zhangii increased whole body lipid level. Dietary autoclaved S. zhangii meal increased gut Lipase and amylase activities, meanwhile those fed 5% raw or autoclaved S. zhangii meal had higher protease activity (P < 0.05). Supplementing diet with S. zhangii meal significantly reduced serum alanine aminotransferase and triglycerides, and enhanced total protein and globulin levels. Liver catalase and superoxide dismutase activities increased by supplementing diet with 5% raw or autoclaved S. zhangii meal (P < 0.05). In addition, catalase activity in fish fed raw S. zhangii meal was higher than those fed autoclaved S. zhangii meal. Blood hemoglobin in fish fed autoclaved S. zhangii meal, especially ASZ2.5 was higher than the other groups. Supplementing diet with 5% raw or autoclaved S. zhangii meal markedly increased respiratory burst, lysozyme and phagocytosis activities. The cumulative mortality against A. invadans infection was 27% and 33%, respectively, when fish were fed diets supplemented with 5% raw or autoclaved S. zhangii meal. Based on the findings of the present study 5% raw S. zhangii meal to feed formulation for L. rohita fingerlings is recommended.
