Liquid extract of Kappaphycus alvarezii enhances growth and metabolic responses in in vitro cultivated explants of this same species

Kappaphycus alvarezii is one of the most important species for seaweed aquaculture but the traditional vegetative propagation methods can reduce crop quality and productivity. This study aimed to evaluate the effects of Kappaphycus Liquid Bioestimulant (KLB) on the growth and metabolic changes of K. alvarezii explants cultured in vitro. Tetrasporophytes from the brown strain of K. alvarezii were cultivated in sterilized seawater enriched with von Stosch nutrient solution (VS50) and subjected to KLB treatments at concentrations of 2%, 4%, and 6% for 14 days followed by another 14 days cultivation only in sterilized seawater enriched with von Stosch nutrient solution (VS50). Explants were analyzed for growth rate, biomass gain, pigment content, and metabolic compounds. Results indicated that KLB treatments significantly increased the number of upright axes, growth rate, and total biomass gain compared to the control. KLB at 4% concentration resulted in the highest growth rate (2.06% day−1) and biomass gain (77.34%). KLB-treated explants also showed enhanced levels of chlorophyll, carotenoids, and total phenolic compounds. Furthermore, KLB application led to a reduction in starch reserves and total soluble sugars, suggesting a shift in energy metabolism towards growth. These findings demonstrate that KLB even at the lowest tested concentration positively influences the regeneration and metabolic activity of K. alvarezii explants, contributing to more efficient and sustainable propagation, highlighting the potential of KLB as a biostimulant for improving K. alvarezii cultivation.
