Enhanced growth and metabolite production in Porphyridium purpureum under nutrient and light treatments

Porphyridium purpureum is one a promising source of valuable substances. In this research, biomass production and the accumulation of bioactive compounds including phycobiliproteins (PBP), carotenoid, chlorophyll and exopolysaccharides (EPS) in P. purpureum were investigated at different concentrations of NaCl and NaNO3 and light treatments (white, red and blue) for 5 weeks in F/2 medium. The best biomass production (0.958 g L-1) was achieved at 50 g L-1 NaCl after 5 weeks of cultivation. The highest content of all pigments was 64.70 mg L-1 PBP (48.92 and 11.24 mg L-1 phycoerythrin (PE) and phycocyanin (PC) respectively), 5.41 mg L-1 chlorophyll a, and 1.90 mg L-1 carotenoids after 4 weeks at 50 g L-1 NaCl and 0.375 g L-1 NaNO3. At the same time, the maximum EPS production (70.97 mg L-1) was achieved in lower concentrations of NaCl (25 g L-1) and NaNO3 (0.075 g L-1). White light acts more efficiently than other lights in biomass (1.23 g L-1) and the content of all pigments (58.42 mg L-1 PBP, 5.81 mg L-1 chlorophyll a, and 1.69 mg L-1 carotenoids) in 4 weeks. Results showed EPS production needs a stationary phase in which the increase occurs after 4 weeks. Also, red light acts as an inducer for EPS enhancement (78.7 mg L-1). External (EPS) and internal (biomass and pigments) metabolite production depends on low concentration of nitrate and higher salinity, respectively.
