Genetic diversity of Brassica oleracea using agromorphological and simple sequence repeat (SSR) markers

As an economically vital crop, cabbage (Brassica oleracea) requires comprehensive characterization of its genetic diversity to optimize its utilization in breeding programs. This study evaluated the genetic diversity of 48 cabbage germplasms using nine agromorphological traits and 20 simple sequence repeat (SSR) markers. The results revealed substantial phenotypic variation among accessions, with genetic diversity indices (H′) ranging from 0.46 to 1.20. Phenotypic clustering partitioned the 48 accessions into five major groups. Among the 20 SSR markers, 18 exhibited polymorphism, amplifying 2–4 fragments per primer (mean = 2.8), yielding a total of 57 polymorphic bands across all accessions. The polymorphic information content (PIC) ranged from 0 to 0.63. Unweighted pair group method with arithmetic mean (UPGMA) clustering based on SSR data classified the accessions into two major clusters. Comparative analysis demonstrated partial congruence between phenotypic and molecular clustering results, with specific accessions showing high similarity in both systems. Integrated phenotypic-SSR approaches provided a more robust assessment of genetic differentiation. Collectively, these findings underscore that the investigated cabbage germplasms represent a valuable reservoir of genetic and agromorphological variability, offering critical resources for future breeding strategies.
