<p><i>Mentha spicata</i> L. (spearmint) is a commercially significant medicinal and aromatic plant, primarily valued for its high carvone content, a monoterpenoid ketone widely employed in the flavor, fragrance, and pharmaceutical industries. Precise identification of <i>M. spicata</i> is vital for ensuring cultivar purity, maintaining genetic integrity in breeding programs, and preventing adulteration in commercial supply chains. However, taxonomic resolution within the genus <i>Mentha</i> is impeded by extensive interspecific hybridization, polyploidy, and morphological convergence. In the present study, a species-specific Sequence Characterized Amplified Region (SCAR) marker was developed for the unambiguous molecular identification of <i>M. spicata</i>. Fourteen Inter-Simple Sequence Repeat (ISSR) primers were screened across six <i>Mentha</i> species, generating 75 polymorphic loci with an average Polymorphic Information Content (PIC) of 0.47, reflecting moderate genetic diversity. Among them, ISSR primer 5 amplified a unique 750&#xa0;bp fragment specific to <i>M. spicata</i> accessions. This species-exclusive fragment was excised, cloned, sequenced, and used to design SCAR-specific primers. The resulting SCAR marker consistently amplified a single 279&#xa0;bp band exclusively in <i>M. spicata</i>, demonstrating high specificity and reproducibility across diverse genotypes. This is the first report of an ISSR-derived SCAR marker for <i>M. spicata</i>, offering a reliable molecular tool for species authentication, germplasm validation, and regulatory compliance. Furthermore, its application in marker-assisted selection will support the development of elite cultivars with elevated carvone content, contributing to both conservation and commercial breeding programs in this economically valuable species.</p> Graphical abstract <p></p>
