Transcriptome sequencing-based genetic diversity assessment of Lagenaria siceraria landraces using simple sequence repeat (SSR) and single nucleotide polymorphism (SNP) markers

Lagenaria siceraria is widely cultivated in various regions of China, where there is a rich diversity of landraces, the fruits of which exhibit significant differences in shape and size. However, the systematic collation of information on the morphological variation and genetic relationships among these landraces are deficiency. In this study, 33 landraces with different fruit shapes were collected, and the phenotypes of fruits, leaves and flowers were systematically classified. Transcriptome sequencing was performed on two landraces with distinct fruit characteristics, ‘Central Asian alvo’ and ‘Manus torquent’, which revealed differences in several pathways, including the biosynthesis pathways of terpenoids and flavonoids, as well as the metabolism of cofactors and vitamins. Based on these transcriptome sequencing data, Simple Sequence Repeat (SSR) and Single Nucleotide Polymorphism (SNP) primers were developed to conduct a systematic classification at the genetic level, trying to elucidate the morphological differences. The results of this study are of great significance for the identification of Lagenaria siceraria landraces and molecular marker-assisted selection.
