<p>Drought stress significantly influences plant growth and improvement by affecting morphology, anatomy, and physiological processes. This study aimed to discover the elite drought-tolerant rice cultivar by screening the landraces collected from various agro-climatic regions of Tamil Nadu. A total of 38 rice landraces, along with two check cultivars, were evaluated under drought using a standardized treatment of − 1.0&#xa0;MPa PEG 6000 under laboratory conditions in the Department of Crop Physiology, Tamil Nadu Agricultural University (TNAU), Coimbatore. Eight landraces showed notable drought tolerance and exhibited superior seedling growth under drought stress. These selected landraces were further analyzed for growth, physiological, and biochemical characterization to assess the effect of drought tolerance under pot culture conditions in the glasshouse. From the pot culture study, six landraces performed better, and they were selected for the field study along with two check cultivars at the Agricultural Research Station, TNAU, Bhavanisagar. Based on this study, Kuliyadichan emerged as the most promising cultivar, followed by Milagusambha was found to excel in all the five morphological traits associated with drought resistance. Regarding physiological characters, Kuliyadichan, Milagusambha, and Rascadam exhibited the highest drought tolerance score. Additionally, the cultivars Kuliyadichan, Milagusambha, Rascadam, and Kallundai showed superior biochemical traits. These same cultivars also offered a higher spikelet fertility and drought tolerance efficiency. Therefore, Kuliyadichan, Milagusambha, and Rascadam are identified as potential candidates for future rice breeding programs aimed at developing drought-resistant cultivars.</p>
