Phylogenetic study of Adenophora koreana by morphological traits and DNA barcoding

Objective and accurate identification of species may be crucial in assessment and maintenance of biodiversity within certain regions. In spite of Adenophora koreana Kitam. 1936 being species endemic to the DPR Korea which was first discovered in Kumgang County, Kangwon Province, it is recorded as homonymous of Adenophora pereskiifolia in the Catalogue of Life (CoL). To elucidate taxonomic uncertainty in the genus Adenophora, principal component analysis-based comparison of morphological traits and DNA Barcode (ITS, psbA-trnH)-based phylogenetic analysis among Adenophora species including A. koreana and A. pereskiifolia were conducted. A. koreana and A. pereskiifolia exhibit significant difference in peduncle length, characteristics of feathers in both leaves and phyllotaxix and in the shape of leaves. Maximum likelihood (ML) dendrogram based on ITS and psbA-trnH distinguished A. koreana and A. pereskiifolia at the genetic distance of 0.002. Morphological and phylogenetic diifferentiation and division in distribution of these two species suggest taxonomic status of A. koreana as endemic species to DPR Korea and requirement of preferential protection, thus contributing to establishment of conservation strategy of genus Adenophora.
