Pollen patterns and morphological variability unraveling taxonomic relevance and evolutionary relationship in salt-tolerant species

Palyno-morphological microscopic study [light microscopy (LM) and scanning electron microscopy (SEM)] of 12 salt-tolerant plant species (Heliotropium curassavicum, Heliotropium europaeum, Euploca strigosa, Capparis decidua, Capparis spinosa, Chrozophora tinctoria, Euphorbia indica, Euphorbia serpens, Alhagi maurorum, Tephrosia purpurea, Polygonum aviculare, and Polygonum plebeium) belonging to five families from the Salt Range was conducted. The main objective was to elucidate the significance of pollen morphology in taxonomy that is beneficial for the identification and discrimination of salt-tolerant taxa. The described morphological pollen characteristics based on habitat are a good source for genera and species differentiation. The systematics of salt-tolerant taxa are strengthened and supported by the examined variation in pollen morphology and the exploration of novel pollen traits. The pollen shape of most of the studied species was suboblate, oblate-spheroidal, subprolate, and prolate-spheroidal, and exine ornamentation was reticulate (7 species) and psilate (3 species). Most species exhibited tricolporate pollen aperture while 6-7-colporate and heterocolpate were found in Chrozophora tinctoria and Heliotropium europaeum. The highest pollen viability values (%) described the adaptability of salt-tolerant taxa in the salt range. Principal component analysis (PCA) and cluster analysis were performed to indicate significant correlations among the species. The pollen morphological features such as aperture, polar and equatorial view, and exine ornamentation are potential taxonomic parameters for species identification and differentiation between the same group taxa.
