<p>Anthropic occupation in watersheds leads to various problems regarding water quality due to the discharge of pollutants. In this context, this study aimed to assess the water quality of the Beberibe River watershed in Northeastern Brazil, aiming to identify possible behaviours and dynamics in areas under different land uses. Data from 2006 to 2018 on dissolved oxygen (DO), biochemical oxygen demand (BOD), ammonia, and phosphorus were used for five monitoring points along the watershed: BE-01, BE-09, BE-30, BE-45, and BE-50. Principal component analysis (PCA), cluster analysis, and Mann–Kendall tests were conducted to study these data's spatial and temporal behaviour. The results indicated significant spatial and temporal variation in water quality, where headwater points exhibited better conditions, with higher DO levels and lower concentrations of nutrients and BOD. In contrast, points near the river's mouth showed a trend of water quality degradation, characterized by increased levels of nutrients and BOD and a reduction in DO. Cluster analysis and PCA revealed a clear distinction between headwater points and others, highlighting the impact of urbanization and agricultural activities on water quality, effectively dividing the watershed into two areas. Correlation analysis showed DO had an inverse relationship with phosphorus ( − 0.73) and ammonia ( − 0.67), while positive correlations among pollutants revealed familiar sources. The Mann–Kendall test detected an increasing BOD trend near the Beberibe River’s mouth (<i>p</i> = 0.042 and <i>p</i> = 0.0185). Therefore, multivariate statistical tools and trend analysis confirmed the relationship between water quality and land use, aiding in identifying priority areas for intervention.</p>
