<p>Anthracnose is a fungal disease that affects all parts of grapevine in warm and humid regions of the world. The present study aimed to assess 13 <i>Vitis</i> spp. germplasm accessions for resistance to grapevine anthracnose, caused by <i>Elsinoe ampelina</i> in southern Brazil, during the 2014/2015 and 2015/2016 growing seasons. Anthracnose incidence and severity were assessed biweekly under natural infection from budburst (October—BBCH-09) until flowering (November—BBCH-63). The accessions were compared using the following epidemiological parameters: (a) beginning of symptom appearance (BSA), (b) time to reach maximum disease incidence and severity (TRMD<i>I</i> and TRMD<i>S</i>), (c) maximum disease incidence and severity (<i>I</i><sub>max</sub>, <i>S</i><sub>max</sub>), and (d) areas under incidence and severity disease progress curves (AU<i>I</i>DPC and AU<i>S</i>DPC). Additionally, histological analyses and quantification of hydrogen peroxide production were conducted on the least and most susceptible accessions to anthracnose. In general, there were significant differences in the estimated epidemiological parameters among accessions. The main parameters that differentiated accessions were <i>I</i>max, <i>S</i>max, AU<i>I</i>DPC, and AU<i>S</i>DPC. <i>Vitis berlandieri</i>, <i>V. shuttleworthii,</i> and <i>V. candicans</i> were the least susceptible accessions to anthracnose, whereas ‘Baco 1’, <i>V. thunbergii,</i> and <i>V. betulifolia</i> were the most susceptible. <i>V. thunbergii</i> and <i>V. betulifolia</i> showed the highest <i>I</i>max, <i>S</i>max, AU<i>I</i>DPC, and AU<i>S</i>DPC values compared with the least susceptible <i>V. berlandieri</i>, <i>V. shuttleworthii,</i> and <i>V. candicans</i>. Additionally, <i>V. betulifolia</i> exhibited greater cell death in leaf tissues around the <i>E. ampelina</i> infected sites than <i>V. shuttleworthii</i>. The variation in susceptibility levels to anthracnose among accessions can be exploited in future grapevine breeding programs.</p>
