Response and evaluation of polyploidy induction on morphology, physiology and drought tolerance of Hydrangea paniculata ‘Pink Diamond’

Hydrangea paniculata ‘Pink Diamond’ (2n = 2x = 36) is a deciduous shrub with high ornamental value, highly favored in commercial floriculture for its long flowering period from summer to autumn and its unique conical inflorescences. This study aimed to establish an efficient protocol for colchicine-induced tetraploidy (2n = 4x = 72) in H. paniculata ‘Pink Diamond’ and to evaluate the morphological, physiological, and drought-resistance characteristics of the tetraploids. Seeds were soaked in colchicine solutions (0.05–0.3%) for 6–30 h in distilled water medium. The highest germination rate (GR, 28.83 ± 2.75%) was achieved after 12 h of treatment with 0.15% colchicine. Chromosome counting, stomatal morphology, and DNA content analysis of greenhouse-grown seedlings confirmed polyploidy induction, with the highest tetraploid induction rate (5.61 ± 1.05%) obtained under 0.2% colchicine treatment for 18 h. Compared with diploids, the induced tetraploids exhibited pronounced morphological and anatomical enhancements: thicker stems (+ 15.67%), larger leaves with improved anatomical structure (leaf thickness + 35.25%, lower epidermis + 15.71%, palisade tissue + 24.58%, spongy tissue + 12.20%, and main vein + 36.66%). Photosynthetic efficiency was markedly improved, with total chlorophyll (Chl) content increased by 30.84% and chlorophyll fluorescence parameter (Fv/Fm) enhanced by 3.24%. Under drought stress, tetraploids maintained higher relative water content (RWC), soluble sugars (SS), soluble proteins (SP), and antioxidant enzyme activities (CAT, POD, SOD), while showing lower accumulation of malondialdehyde (MDA) compared with diploids. This study demonstrates that tetraploid H. paniculata ‘Pink Diamond’ possesses greater growth vigor, photosynthetic capacity, and drought tolerance, providing valuable variant materials and technical support for the breeding of new H. paniculata cultivars.
