Development and characterization of Aegilops tauschii Coss. pau14232 derived hexaploid wheat introgression lines exhibiting resistance to bird cherry-oat aphid, Rhopalosiphum padi L.

A set of ten aphid-resistant BC2F3 hexaploid wheat introgression lines (IL 36, 39, 45, 51, 69, 100, 104, 105, 111, and 112) were developed from the cross of Triticum durum cv. PBW114/Aegilops tauschii acc. pau14232//3*BWL4478. Investigations on different categories of host plant resistance (HPR) against bird cherry-oat aphid (BCOA), Rhopalosiphum padi L., were carried out on ILs along with Ae. tauschii acc. pau14576 (diploid resistant), Ae. tauschii acc. pau14109 (diploid susceptible), Ae. tauschii acc. pau14232 (resistant parent) and hexaploid cultivar HD 2967 (susceptible parent), in winter cropping season (November to April), 2021–22. No-choice experiments were conducted in screen house to study the locomotory behavior, nymphiposition preference, fecundity, intrinsic rate of increase (rm), mean relative growth rate (mRGR), and survival of BCOA. R. padi moved less on IL39 (0.2 ± 0.01) and quickly found feeding site, while IL104 (0.48 ± 0.05) was the least preferred host. Introgression line 36 showed the least nymphiposition preference, with a total fecundity of 32 ± 0.89 nymphs. In contrast, IL 51 recorded the highest fecundity of 36.2 ± 0.8 nymphs. IL 36 was more resistant, with fewer nymphs and lower survival. Varied defense responses were observed among different ILs. Analysis showed fecundity was positively correlated with mRGR, weight gain and negatively correlated with nymphal duration. This study found that the R. padi resistance in Ae. tauschii is largely of antibiotic type with Ae. tauschii derived ILs, showing a marginal difference in antixenosis and tolerance studies. The material developed and information generated in this study will/could help develop a high-yielding BCOA-resistant hexaploid wheat cultivar.
